Vígjátékaiban megformált karakterei látszólag esetlennek, szórakozottnak tnnek, és mindehhez társulnak a filmjeit jellemző abszurd helyzetek (amik főleg a háttérben zajlanak).

Kezdő színészként játszotta komolyabb szerepeit. Főszerepet játszott az 1956-os Tiltott Bolygó cím science fiction-ben, valamint a "Poszeidon-katasztrófa" cím filmben, mint hajóskapitány. Ezután jónéhány év múlva fordult csak a vígjátékok felé. Az 1980-as Airplane!-ben "pléhpofájú" doktorként nyújtott alakítása nagyot lendített karrierjén. Egy neves filmkritikus, Roger Ebert mondta róla: "A svindlik Olivier-je".

Korai élete:
Nielsen a kanadai Regina, Saskachewan-ban született Maybelle és Ingvard Nielsen fiaként. Nielsen édesanyja walesi, míg édesapja dán származású. Két bátyja van, közülük Erik Nielsen a 80-as években Kanada miniszterelnök helyettese volt, unokaöccse, Jean Hersholt később színész lett. Nielsen kipróbálta magát különböző szerepekben, amik sosem érték el Frank Drebin sikerét. Több film is készült a Csupasz Pisztoly trilógia stílusában, változó kritikai és közönségi sikerekkel. Akadtak filmek, melyeket a kritikusok nem kíméltek szegényes teljesítményük miatt.Mivel a Csupasz pisztoly sorozat egy rendőrdráma paródia, Nielsen a későbbiekben is filmparódiákra koncentrált. Rossz kritikát kapott a "Bújj, bújj, ördög!" Magánélet:
Nielsen négyszer nősült, első három felesége: Monica Boyer (1950-1956), Alisande Ulmann (1958-1973), és Brooks Oliver (1981-1983). 2001-ben vette el negyedik feleségét, Barbaree Earlt, akivel már 1983 óta alkottak egy párt.
